DESCRIPTION:Nuclear architecture coordinates patterns of gene expression a
nd mediates cell identity and state. Nuclear architecture and shape are al
so fundamentally altered in most cancers and these abnormalities are routi
nely used to diagnose cancer and stage its aggressiveness. However\, the c
auses and direct consequences of nuclear shape alterations in both healthy
and diseased states have remained unclear. Using chromosome mis-segregati
on driven aneuploidy and colorectal cancer (CRC) as models we have study t
he roles of nuclear mechanics and deformation in cell state regulation\, m
alignant transformation and cancer progression. In my presentation I will
discuss our recent research on how cells sense nuclear deformation and me
chanical changes and how these mechanosignals are integrated with biochemi
cal inputs to alter cell behaviors and tune disease outcome.
